UJI AKTIVITAS INHIBITOR ENZIM XANTIN OKSIDASE
EKSTRAK ETIL ASETAT JAMUR ENDOFIT
DARI KULIT BUAH RAMBUTAN (Nephelium lappaceum L.)
ABSTRAK
Hiperurisemia adalah kondisi dimana terjadi peningkatan kadar asam urat
diatas normal sehingga dapat menyebabkan penumpukan kristal asam urat di
jaringan. Xantin oksidase adalah enzim yang berperan dalam mengkatalisis
oksidasi hipoxantin menjadi xantin dan asam urat. Pada tanaman rambutan (
Nephelium lappaceum L.) telah ditemukan senyawa yang berperan sebagai
inhibitor enzim xantin oksidase yaitu Tanin, flavonoid, polifenol dan asam ellagat.
Pemanfaatan jamur endofit termasuk salah satu mencegah kelangkaan. Jamur
endofit mampu menghasilkan senyawa metabolit sekunder yang sama dengan
tanaman inangnya karena adanya transfer genetik. Tujuan dari penelitian ini yaitu
untuk mengetahui pengaruh ekstrak etil asetat jamur endofit dari kulit buah
Rambutan (Nephelium lappaceum L.) dalam menghambat enzim xantin oksidase.
Penelitian ini menggunakan metode ekstraksi maserasi, dengan berat isolat jamur
endofit 35,50 gr, berat ekstrak 2,121 gr, dan rendemen ekstrak 5,97%. Data hasil
penelitian didapatkan pada persen inhibisi dan IC50 ekstrak etil asetat pada
konsentrasi 125 nilai inhibisinya (52,694%), konsentrasi 62,5 nilai inhibisinya
(32,660%), 31,25 (7,239%), 15,62 (3,620%), 7,81 (2,946%) dengan nilai IC50
sebesar 116 μg/mL. Sehingga, dapat diketahui bahwa ekstrak etil asetat jamur
endofit dari kulit buah Rambutan (Nephelium lappaceum L.) berpotensi dalam
menghambat aktivitas enzim xantin oksidase yang mana ditunjukkan nilai serapan
yang semakin kecil.

ENDOPHYTIC FUNGI ETHYL ACETATE EXTRACT
FROM RAMBUTAN SKIN (Nephelium lappaceum L.)
ABSTRACT
Hyperuricemia is a condition where there is an increase in uric acid levels
above normal so that it can cause a buildup of uric acid crystals in the tissues.
Xanthine oxidase is an enzyme that plays a role in catalyzing the oxidation of
hypoxanthine to xanthine and uric acid. In the rambutan plant (Nephelium
lappaceum L.) compounds have been found that act as inhibitors of the xanthine
oxidase enzyme, namely tannins, flavonoids, polyphenols and ellagic acid. The
use of endophytic fungi is one way to prevent scarcity. Endophytic fungi are able
to produce secondary metabolites that are the same as their host plants due to
genetic transfer. The purpose of this study was to determine the effect of the ethyl
acetate extract of the endophytic fungus from the rind of the rambutan fruit
(Nephelium lappaceum L.) in inhibiting the xanthine oxidase enzyme. This study
used the maceration extraction method, with an endophytic fungus isolate weight
of 35.50 g, an extract weight of 2.121 g, and an extract yield of 5.97%. The
research data obtained on the percentage of inhibition and IC50 of ethyl acetate
extract at a concentration of 125 inhibition values (52.694%), concentrations of
62.5 inhibition values (32.660%), 31.25 (7.239%), 15.62 (3.620%), 7 .81
(2.946%) with an IC50 value of 116 μg/mL. Thus, it can be seen that the ethyl
acetate extract of the endophytic fungus from the rind of the rambutan fruit
(Nephelium lappaceum L.) has the potential to inhibit the activity of the xanthine
oxidase enzyme which shows a lower absorption value.
